Physiological Responses of Grapevines to Waterlogging and Injury Reduction
Abstract
침수가 포도나무에 미치는 피해 양상을 파악하고 용존산소 공급과 착과가 포도나무의 침수 피해 경감에 미치는 영향을 구명하기 위하여 2005년과 2006년에 걸쳐 포트에 심은 2년생 켐벨얼리와 거봉 품종에 5~6주간 침수 처리를 한 후 생육, 양분 흡수 및 광합성 특성과 엽록소 형광 반응 등을 조사하였다.
침수 기간이 길어질수록 포도 유목의 광합성(ACO₂), 기공전도도(gs), 세포내 CO₂농도(Ci), 증산율(E)은 감소하는 반면 엽온을 기준으로 한 대기와 엽간의 확산압차(VpdL)는 높아졌다. 침수시작 후 14일부터 포도 잎은 침수 스트레스를 심하게 받아 제2 광계의 최대광량자수율(Fv/Fm), 양자수율(yield) 및 전자전달율(ETR)이 낮아졌다.
침수에 의해 유엽과 성엽 모두 엽록소 함량이 감소하였고, 엽온이 상승하여 대기와 엽온 간의 온도차가 줄어들었으며, 엽수분포텐셜이 낮아졌다. 초장, 엽수, 줄기 직경, 생체중과 건물중이 감소하였고 T/R율은 침수 기간이 길어질수록 높아졌다. 과실은 착색이 빨라졌으며 일정 기간 침수 후 배수 처리를 했을 경우에는 침수 기간이 길었던 처리구일 수록 당함량이 높고 산함량이 낮아지는 경향을 나타내었다. 하지만 계속 침수상태로 유지된 포도는 당함량이 다소 낮아지는 경향이었다.
침수 후 뿌리조직 내에서는 피층(cortex)내 세포가 융합하여 통기조직(aerenchyma)이 발달되고 물관부 조직이 확대되는 현상이 관찰되었다.
캠벨얼리가 거봉에 비하여 침수 기간 동안 광합성이 다소 높게 유지되고 대기와 엽온 간의 온도차도 낮게 유지되는 것으로 보아 침수 내성이 다소 강한 것으로 판단되었다. 캠벨얼리에서의 침수 피해 발생 임계점을 찾기 위해 침수 기간을 달리하였을 때 1~3일간의 침수는 포도 생육에 큰 영향을 미치지 않았으나 침수 후 7일째부터는 생체중과 건물중이 감소되었다.
침수 피해 경감을 위한 재배적 방법으로서 침수 시에 용존산소를 공급한 결과 침수에 의한 잎의 엽록소함량, ACO₂, gs, E, Fv/Fm과 ETR의 감소가 완화되었으며 VpdL의 증가가 줄어들었다. 또한 용존산소공급에 의해 엽온 상승과 엽수분포텐셜의 감소가 완화되었고 뿌리활력이 향상되었다. 수체의 무기물 흡수는 용존산소공급에 의해 줄기는 P, K, Mg, Na 잎은 P, Ca, Mg, Mn과 엽병은 P, Ca, Mg, Fe, Zn 함량이 침수구에 비해 증가되었다.
캠벨얼리 품종에서 침수되지 않은 정상토양 조건에서는 비착과구가 착과구에 비해 광합성과 생육량(초장, 엽수, 생체중, 건물중)이 많은 반면, 침수 조건에서는 착과된 포도나무가 광합성과 생육량(초장, 엽수, 생체중, 건물중)이 많았다. 4주간 침수 후 배수 처리를 하였을 때 침수에 의한 광합성 저하는 착과구에서 빨리 회복되는 경향을 나타내었으며 뿌리활력도 높았다. 또한 2주간 침수 조건에서 수체의 무기물 흡수는 착과에 의해 잎은 N, P, K, Zn, 엽병은 N, K, Zn, 뿌리는 N, P, K, Ca가 증진되었다.
결론적으로 포도 켐벨얼리 품종에서 침수에 의해 피해를 받는 한계 침수 기간은 3일과 7일 사이에 있으며, 침수 조건에서 용존산소 공급과 착과가 포도나무의 뿌리활력, 무기양분의 흡수, 광합성 개선을 통하여 생육을 좋게 하고 침수 스트레스를 감소시켜 포도나무의 침수 피해를 경감시켰다.
This experiment was conducted to elucidate the damage of waterlogging, and the effect of supplying dissolved oxygen and fruiting to waterlogged grapevine. Growth and characteristics of nutrient uptake, photosynthesis, and chlorophyll fluorescence were investigated for potted two-year-old waterlogged grapevine in 2005 and 2006.
Longer period of waterlogging reduced net CO₂ assimilation (ACO₂), conductance to H₂O (gs), intercellular CO₂ concentration (Ci) and transpiration rate (E) and increased leaf-to-air water vapor pressure deficit on leaf temperature (VpdL). Maximum quantum yield of photosystem Ⅱ (Fv/Fm), electron transport rate (ETR), and yield of grapevine leaves were decreased from 14 days after waterlogging.
Waterlogging stress resulted in the decrease of chlorophyll contents of young and mature leaves, and temperature difference between leaf and air was diminished. Prolonged waterlogging decreased leaf water potential significantly. It also reduced the shoot length, the number of leaf, stem diameter, fresh and dry weight of grapevine due to inhibiting root formation and hypoxia around roots. Meanwhile, T/R ratio increased with prolonged waterlogging. On the other hand, waterlogging enhanced the development of fruit color. Draining after a given period of waterlogging increased soluble solid contents but decreased titratable acid contents. Soluble solid contents of grapes decreased with prolonged waterlogging without draining.
Microscopic observation showed that root tissues of waterlogged grapevine developed aerenchyma tissue by disintegrating cortex cells and enlarging xylem tissues.
Higher net CO₂ assimilation and smaller difference between leaf and air temperature in 'Campbell Early' than in 'Kyoho' during the waterlogging period indicated that 'Campbell Early' is more tolerant to waterlogging. One to three days of waterlogging did not affect the growth of grapevine: fresh and dry weight of grapevine decreased after seven days of waterlogging, indicating that the critical period of waterlogging in 'Campbell Early' is shorter than seven days.
Supplying dissolved oxygen to waterlogged grapevine alleviated the decrease of chlorophyll contents, ACO₂, gs, E, Fv/Fm, ETR as well as the increase of VpdL. It also increased root activity, difference between leaf and air temperature, and alleviated decreasing of leaf water potential. Besides, it increased uptake of P, K, Mg, and Na in stems, P, Ca, Mg, and Mn in leaves, and P, Ca, Mg, Fe, and Zn in petioles.
Decreased CO₂ assimilation and growth were observed in fruited 'Campbell Early' compared to those of non-fruiting trees under non-waterlogged condition. Under waterlogged conditions, net CO₂ assimilation, and growth (shoot length, leaf number, fresh and dry weight) in fruiting trees were improved compared to non-fruiting.
Draining after four weeks of waterlogging showed that 'Campbell Early' fruiting trees recovered more quickly and had higher root activity than non-fruiting trees. It also improved the uptake of mineral nutrients in leaves (N, P, K, and Zn), petioles (N, K, and Zn), and root (N, P, K, and Ca) of 'Campbell Early' grapevine after a two-week waterlogging period.
In conclusion, the critical period of waterlogging without any injury in 'Campbell Early' was between three and seven days. Supplying dissolved oxygen and fruiting on waterlogged grapevine increased the tolerance to the stress and alleviated the inhibition of growth by improving the root activity, the photosynthesis, and the uptake of mineral nutrients.
