Nutritional and immunogical studies of marine fish fed Bacillus spp. supplemented diets

Abstract
양식산업은 식량산업에서 중요한 분야로 부각되어 있으며 지속적인 성장을 보이고 있다. 하지만 최근들어 고밀도 양식과 무분별한 항생제의 사용으로 인한 내성균의 발생으로 어류질병의 치료는 더욱 어려워 경제적으로 막대한 손실을 초래하고 있다. 이러한 문제를 해결하기 위해 어류의 성장과 면역능력을 향상시키는 사료첨가제, 면역증가제의 개발에 관한 연구가 수행되고 있으나 일반적으로 그 효능은 미비한 실정이다. Probiotic (생균제)의 어원은 그리스어로 "For life"이며 probiotic을 적정 공급하였을 때 숙주의 장내 건강을 향상시킬 수 있는 살아있는 미생물로 정의를 내린다. 양어사료에서 probiotic은 성장촉진제, 친환경 항생제로 사용이 가능하며 양어사료 내 probiotic의 첨가는 여러 어종에서 성장촉진, 사료효율 및 면역력 증강에 따른 질병저항성을 강화 시킬 수 있는 첨가 물질로 보고 되고 있다. 따라서 본 연구는 최근 각광받고 있는 Bacillus spp. probiotic의 첨가가 국내 대표적인 해산 양식어류인 넙치와 참돔의 성장, 사료효율, 면역활성 및 여러 질병균에 대한 질병저항성에 어떠한 영향을 미치는지를 조사하기 위해 수행되었다.

본 학위논문 연구에서는 일련의 연구들을 통해 Bacillus 균주 probiotic의 이용 가능성을 총 3가지의 실험연구를 통해 입증하고자 하였다. 연구 1에서는 (Chapter II) 치어기의 넙치를 대상으로 여러 종류의 Bacillus 균주들을 평가하여 이용 가능성이 가장 높은 균주를 선발하였으며, 연구 2 (Chapter III)에서는 연구 1의 실험 결과들을 통해 선별된 유용균주인 Bacillus subtilis를 사료 내 농도별로 첨가하여 B. subtilis의 이용성과 적정 첨가함량을 규명하였다. 연구 3 (Chapter IV)은 참돔을 대상으로 B. subtilis + B. licheniformis의 혼합 첨가와 첨가 농도에 따른 Bacillus 균주들의 이용 가능성 및 그 시너지효과를 알아 보기 수행되었다.

연구 1의 결과, 사료 내 B. subtilis 균주의 첨가는 넙치 치어에 있어서 성장률, 사료효율, 단백질전환효율, 비특이적 면역력, Streptoccocus iniae에 대한 질병저항성을 높일 수 있는 것으로 나타났다. 다른 종류의 균주(B. pumilus & B. licheniformis)는 향상효과가 있었지만 유의적이지는 못했다. 연구 2에서는 B. subtilis 균주를 넙치 사료 내 농도별(0.1%, 0.3%, 0.5%)로 첨가하여 실험 한 결과, 성장률, 사료효율, 단백질이용효율 및 비특이적 면역력에서 0.5%의 B. subtilis를 첨가한 실험구에서 유의적으로 높은 결과를 보였다. 또한, 적정량의 사료 내 B. subtilis의 첨가는 어류의 장 내 유용균주의 증가에도 중요한 역할을 하는 것으로 나타났다. S. iniae에 대한 질병저항성 실험 결과에서도 B. subtilis를 첨가한 실험구에서 대조구에 비해 유의적으로 높은 생존율을 보임으로써, 사료 내 B. subtilis의 첨가는 넙치의 질병저항성에도 매우 중요한 역할을 할 것으로 판단되었다. 연구 3에서는 성장율과 사료효율 등에서 대조구에 비해 첨가 실험구들이 유의적인 차이를 보이지 않았으나 혼합균주를 사용한 모든 실험구에서 대조구에 비해 어류의 스트레스를 유의적으로 감소시킬 수 있는 것으로 나타났다. 비특이적 면역력과 S. iniae에 대한 질병저항성에서는 혼합균주의 첨가가 유의적으로 높게 나타났지만 이에 대해서는 보다 구체적인 연구가 필요할 것으로 사료된다.

위 결과들을 종합해 볼 때, 사료 내 Bacillus spp.의 첨가는 대표적인 해산 어류인 넙치와 참돔에서 성장과 사료효율을 증대시킬 수 있을 것으로 보이며, 어류 사육시의 스트레스 감소로 인한 어류의 생리활성 촉진으로 비특이적 면역력의 증진과 그에 따른 어류의 질병저항성도 높일 수 있을 것으로 사료된다. 하지만 보다 구체적이고 지속 가능한 결론을 도출하기 위해서는 타 어종에서의 장기간의 첨가 실험과 실험환경을 달리한 현장 실험에서의 적용 등 후속 연구들이 뒷받침 되어야 할 것으로 사료된다.
Olive flounder is currently the most important marine fish cultured in Korea. It is expected that the demand for aquatic food will increase because of the increase in the human population and the perception that aquatic food is healthy. However, past several decades, this industry has a high density stocking strategy in the pursuit of maximum productivity in limited culture systems. As a result, intensive aquaculture triggers higher susceptibility to disease and significant losses. Researchers have focused on finding potential alternatives for antibiotics due to the negative effects of using antibiotics for controlling fish disease, including outbreaks of antibiotic resistant bacteria and concerns about environmental hazards and food safety. Their utilization in aquaculture practices needs to be restricted due to the occurrence of antibiotic-resistant bacterial strains, water pollution in the aquaculture environment and accumulation of residual antibiotics in aquaculture products.
As an environment-friendly measure of controlling fish disease, probiotics have been recently received great attention in aquaculture industry. A wide range of probiotics have been examined in aquaculture that most of them belong to Saccharomyces, Clostridium, Lactobacillus, Bacillus, Enterococcus, Shewanella, Leuconostoc, Lactococcus, Carnobacterium and Aeromonas species. Extensive studies on probiotics have demonstrated that they are not only an efficient replacement for antibiotics in aquaculture but also possess numerous positive effects, including enhanced fish growth, improved feed efficiency through the production of exoenzymes, modulation of intestinal microflora, enhanced immune response, antagonism to pathogens. The immunomodulatory activity of probiotics is a key factor for preventing fish disease. However, the exact mechanism by which probiotics exert such effects is still unclear.
Therefore, we investigated the effects of different levels of dietary Bacillus spp. for olive flounder and red seabream to establish an optimal dietary inclusion level in terms of innate immunity, disease resistance and growth performance.

The results indicated that probiotic, such as Bacillus spp (1010). Supplements could replace antibiotic without negative effects on growth performances, feed utilizations, innate immunity and survival of juvenile olive flounder (Chapter II). Dietary supplementation of B. subtilis can increase growth performance, feed utilization, innate immunity and disease resistance of juvenile olive flounder against S. iniae. The optimum dietary level seems to be B. subtilis (1010) for olive flounder (Chapter III). Even though there was no significant different growth performance, dietary supplementation of mixed Bacillus spp. (B. subtilis and B. licheniformis) can increase disease resistance of red seabream against S. iniae and eventually reduce its overall mortality (Chapter IV).
