Abstract
Nojirimycin (NJ) is a structure in which the oxygen of the ring is replaced with an NH group in the D-glucose structure, and 1-deoxynojirimycin (DNJ) is produced when NJ is dehydrated and reduced. NJ, which have similar structures to D-glucose, are powerful glucosidase inhibitors and are interesting compounds. However, no anti-inflammatory effect has been reported. Therefore, in order to investigate anti-inflammatory effect, the production and expression of inflammatory cytokines as well as inflammatory mediators such as iNOS and COX-2 were measured in LPS-stimulated RAW264.7 macrophages. In addition, the mechanisms of representative inflammatory signaling pathways, suppression of NF-κB (nuclear factor-kappa B) and MAPKs (mitogen activated protein kinase) activation were studied. The production of iNOS, COX-2, and inflammatory cytokines (PGE2, IL-6, IL-1β, TNF-α) according to NJ and DNJ treatment was significantly inhibited. In addition, DNJ showed anti-inflammatory effect by suppressing LPS-induced MAPKs activation, and NJ, DNJ showed anti-inflammatory effect by suppressing the translocation of NF-κB through the prevention of IκBα (inhibitory factor kappa B alpha) phosphorylation. In addition, only NJ showed whitening effect, which is implied due to structural differences. Summarizing the above results, there is a possibility that NJ and DNJ can be used as anti-inflammatory agents, and it is implied that NJ can also be used as a whitening agent.
